Resumen

RESUMEN: El cultivo de cítricos es afectado por varios hongos fitopatógenos, los cuales pueden causar diversas enfermedades y reducir su producción. El hongo Lasiodiplodia spp. causa muerte regresiva y necrosis en cítricos y en una amplia gama de cultivos en diversos países, incluyendo México. Los agentes microbianos antagónicos pueden controlar el desarrollo y la propagación de los patógenos mediante diversos mecanismos. El objetivo de este trabajo fue aislar e identificar mediante técnicas tradicionales y moleculares los hongos presentes en árboles de cítricos causando necrosis y muerte regresiva en huertas de Nuevo León y Tamaulipas; así como también aislar y evaluar el antagonismo de microorganismos nativos del suelo contra los hongos fitopatógenos causantes de dicha enfermedad. Los hongos aislados fueron identificados con base en sus características morfológicas como Lasiodiplodia theobromae, Fomitopsis meliae y Eutypella citricola, los cuales fueron confirmados mediante la secuenciación del ADN de la región ITS y su comparación con secuencias en el GenBank. En el bioensayo de invernadero se comprobó que L. theobromae y F. meliae causaron marchitez, muerte descendente y lesiones necróticas en las áreas inoculadas a partir del cuarto día posterior a la inoculación. E. citricola sólo produjo necrosis alrededor del área inoculada. F. meliae + L. theobromae causaron daños más severos, provocando lesiones necróticas de 22 a 27 cm de longitud, marchitez y muerte regresiva, y ambos fueron re-aislados a partir de las lesiones producidas. De acuerdo con los resultados obtenidos, los hongos L. theobromae y F. meliae causan muerte regresiva de los cítricos. Adicionalemente se logró el aislamiento de 15 cepas microbianas con actividad antagónica, y tres de ellas, el actinomiceto M4 R, el hongo M104 (Penicillium citrinum) y el actinomiceto M1-101 mostraron porcentajes de inhibición del 84, 85 y 91% respectivamente contra L. theobromae. En la prueba de inhibición de la germinación, se observó la producción de clamidosporas como un medio de supervivencia de los patógenos cuando están frente a algún compuesto antifúngico o microorganismo biocontrolador. Los fungicidas dicarboximida, oxicloruro de cobre + clorhidrato de oxitetraciclina + sulfato de estreptomicina, y cloratolonil + cymoxanil a las concentraciones de 3, 0.6 y 3.5 µg mL-1 respectivamente fueron incapaces de reducir el crecimiento de L. theobromae en las condiciones evaluadas, sin embargo, el fungicida clorotalonil + cymoxanil logró inhibir el crecimiento de F. meliae en un 50%. ABSTRACT: Citrus crops are affected by several phytopathogenic fungi, which can cause various diseases and reduce their production. The fungus Lasiodiplodia spp. causes dieback and necrosis in citrus and in a wide range of crops in various countries, including Mexico. Antagonistic agents could controling the development and spread of pathogens by different mechanisms. The aim of this work was to isolate and identify the fungal species obtained from citrus trees with symptoms of dieback from Nuevo Leon and Tamaulipas groves, and to isolate and evaluate native soil microorganisms with antagonistic capacity against pathogenic fungi causing that disease. The isolated fungi were identified based on their morphological characteristics such as Lasiodiplodia theobromae, Fomitopsis meliae and Eutypella citricola, confirmed with DNA sequencing of the ITS region when was compared with sequences in GenBank. In the greenhouse bioassay, it was found that L. theobromae and F. meliae caused symptoms of wilting, descending death and necrotic lesions in the inoculated areas since the fourth day after inoculation. E. citrícola only produced necrosis around the inoculated area. F. meliae + L. theobromae caused more severe damage, such as necrotic lesions from 8.66 to 10.63 in. in lenght, wilting and dieback. Both were re-isolated from the lesions produced. According to the results obtained, the fungi L. theobromae and F. meliae cause citrus dieback. Aditionally, isolation of 15 strains with antagonistic activity was obtained, and among them, three (M4 R actinomycete, M104 Penicillium citrinum and M1-101 actinomycete) showed percentages of inhibition of 84, 85 and 91% respectively against L. theobromae. In the germination inhibition test, the production of chlamydospores was observed as a means of survival of the pathogens when they are against an antifungal compound or biocontrol microorganism. The fungicides dicarboximide, cuprum oxichloride + oxitetracycline chlorhidrate + streptomycine sulphate, and cloratolonil + cymoxanil to 3, 0.6 y 3.5 µg mL-1 concentrations respectively were unable to reduce the growth of L. theobromae, however the fungicide chlorothalonil + cymoxanil was able to inhibit F. meliae growth by 50%.
