Resumen

Se aislaron e identificaron compuestos con actividad antimicrobiana en contra de una cepa farmacorresistente de Acinetobacter baumannii, esto a partir de extractos biológicamente activos de Parthenium incanum y Nothoscordum bivalve. Para ello, se llevaron a cabo diferentes extracciones del material vegetal, obteniendo 4 diferentes extractos por planta. En la actividad antimicrobiana preliminar, sólo los extracto metanólicos mostraron efecto. En la obtención de la CMB, se registraron valores de entre 7.50±0.54 y 8.50±0.54 mg/mL. Para la letalidad por Artemia salina, se observó que los extractos presentaron una DL50 de entre 380 y 1882 ppm. En la citotoxicidad preliminar, los extractos causaron entre el 0 y el 92.98% de hemólisis. En la protección de los eritrocitos frente al radical AAPH, los extractos registraron una citoprotección de hasta 94.58%. Con respecto a la captura de radicales, se obtuvieron valores de entre 275.55±21 y 598.99±4 µmoles quivalentes de Trolox/g de extracto. Para los Fenoles Totales Solubles se determinaron valores de entre 1.35±0.07 y 4.13±0.04 g equivalentes de ácido gálico por cada 100 g de extracto. A los extractos de menor letalidad y citotoxicidad, se les determinó el efecto de las concentraciones sub-letales de las CMB´s, sobre el crecimiento en A. baumannii, demostrando que estás no afectaban el crecimiento, pero si la formación de su biopelículas hasta un 41.62%; por otro lado, se estudió la actividad coagulante, determinado que el extracto de P. incanum afectó el TP de manera significativa (p<0.05). En el extracto seleccionado de N. bivalve, se evaluó su efecto sobre la expresión génica de la biopelícula en A. baumanni y se realizó un análisis por MEB; determinando que el extracto afectó la expresión de los genes abaI, bap y csuE, causando una sobreexpresión de 15.4 veces, así mismo, al observar la por microscopia se logró apreciar un daño en la estabilidad y formación de su biopelícula. Así mismo, se le evaluó su citotoxicidad con células normales Pbmc´s, encontrando que hasta 500 ppm no causaba daño en su proliferación celular. Estudios de la inhibición de la enzima α-glucosidasa mostró que el extracto posee una CI50 de 1393±13 ppm y su capacidad antiurolítica mostró un valor de CI50 de 1141±30 ppm. En el caso de P. incanum se aislaron e identificaron la partenina y la coronopolina. Para N. bivalve se identificaron de manera parcial amidas o aminas por FT-IR del extracto sometido a todas las evaluaciones biológicas antes mencionadas. ABSTRACT They were isolated and identified compounds with antimicrobial activity against a drugresistant strain of Acinetobacter baumannii, this from biologically active extracts of Parthenium incanum and Nothoscordum bivalve. To do this, different extractions of the plant material were carried out, obtaining 4 different extracts per plant. In the preliminary antimicrobial activity, only the methanolic extract showed effect. In obtaining the CMB, values between 7.50±0.54 and 8.50±0.54 mg/mL were recorded. For lethality by Artemia salina, it was observed that the extracts presented an LD50 of between 380 and 1882 ppm. In the preliminary cytotoxicity, the extracts caused between 0 and 92.98% of hemolysis. Protection of erythrocytes against the radical AAPH, the extracts registered a cytoprotection of up to 94.58%. With respect to the capture of radicals, values of between 275.55±21 and 598.9λ±4 ȝmol of quivalent Trolox/g extract were obtained. For Total Phenols Soluble, values between 1.35±0.07 and 4.13±0.04 g gallic acid equivalent per 100 g of extract were determined. To the extracts of lower lethality and cytotoxicity, the effect of the sub-lethal concentrations of the CMB's, on the growth in A. baumannii was determined, showing that they were not affecting the growth, but the formation of their biofilms until 41.62%. On the other hand, the coagulant activity was studied, determined that the extract of P. incanum affected TP significantly (p <0.05). In the selected extract of N. bivalve, its effect on the gene expression of the biofilm in A. baumanni was evaluated and an analysis was performed by SEM; determining that the extract affected the expression of the genes abaI, bap and csuE, causing an overexpression of 15.4 times, likewise, when observing the microscopy it was possible to appreciate a damage in the stability and formation of its biofilm. Likewise, its cytotoxicity was evaluated with normal Pbmc's cells, finding that up to 500 ppm did not cause damage in its cellular proliferation. Studies of the inhibition of the enzyme α-glucosidase showed that the extract has an IC50 of 1393±13 ppm and its anti-urolithic capacity showed an IC50 value of 1141±30 ppm. In the case of P. incanum, parthenin and coronopolin were isolated and identified. For N. bivalve, amides or amines were partially identified by FT-IR of the extract subjected to all the aforementioned biological evaluations.
