Resumen

El cáncer cérvico-uterino da muerte a una gran cantidad de mujeres a nivel mundial sin mencionar que en México ocupa el primer lugar de muerte por cáncer. La aparición del cáncer cervical ha sido asociada, en 99% de los casos, con la infección previa del virus de papiloma humano (HPV) en especial los serotipos 16 y 18. La actividad carcinógena del HPV se debe, principalmente, a la integración de DNA viral en el genoma de la célula hospedera y la expresión constitutiva de los genes E6 y E7, cuyos productos peptídicos interfieren con las proteínas endógenas P53 y Rb que son vitales en la vía de supresión de tumores. Se ha demostrado que la expresión continua de las onco-proteínas E6 y E7 es necesaria para la conservación del estado maligno de las células transformadas y que la represión de alguna de ellas puede inducir arresto en el crecimiento o apoptosis en las células carcinógenas. Se ha encontrado, además, que la vía de Mdm2, que es el represor natural de p53, se encuentra aparentemente funcional pero al mismo tiempo sustituida por E6. Es posible que al reprimir la expresión de E6, la vía de Mdm2 vuelva a estar funcional, más no ha sido reportado hasta el momento. Se ha logrado reprimir de manera eficiente y por diversos métodos la expresión de la proteína E6 más nunca se ha comprobado que efecto pudiera tener esta supresión sobre la expresión del gen mdm2. Se han desarrollado diferentes estrategias para la supresión de genes, entre las cuales se encuentra integrada, de manera relativamente reciente, el uso del RNA de interferencia (RNAi). El efecto del RNAi consiste en el silenciamiento de genes mediante la degradación específica del RNA mensajero correspondiente a la proteína que se desea suprimir, tal efecto de silenciamiento puede alcanzar hasta un 85% de efectividad y es conocido como knock-down. Tal eficiencia y especificidad de la vía de silenciamiento post-transcripcional lo vuelve una estrategia importante en la búsqueda de los efectos que pudieran presentarse en la expresión de un gen al silenciar otro que pudiera estar relacionado. En este trabajo se silenciaron los mRNA de E6 y E7 (HPV-16) por medio de RNAi y se analizó, por medio de PCR semicuantitativo, el efecto que el silenciamiento de alguno de estos oncogenes tiene sobre la expresión de mdm2 encontrando que la modificación en la expresión de E6 y E7 no tiene efecto alguno sobre mdm2, sin embargo, al inhibir la expresión de mdm2 el patrón de expresión de E6 y E7 se ve modificado en forma negativa mientras que la expresión de p53 se ve alterada en forma positiva. Abstract Cervical cancer is currently one of major causes of death amongst women world wide, and in Mexico is the main cause of death by cancer. The incidence of cervical cancer has been greatly associated, in as much as 99% of the cases, with the previous infection of human papilloma viruses, particularly the serotypes 16 and 18. The carcinogenic potential of HPV infections is mainly due to the integration of the viral DNA into the genomic material of the host cell and the subsequent constitutive expression of the E6 and E7 genes, which have the ability of repressing tumor suppressing proteins such as P53 and Rb. It has been proved that the continuous expression of these oncoproteins is essential for the conservation of the malignant state of the transformed cells and that the repression of either, can and will induce growth arrest or apoptosis or both. Moreover, it is important to notice that though Mdm2, P53 natural repressor, is apparently functional, it is at the same time, completely replaced by E6. It is possible that in repressing E6, Mdm2 will quickly resume its normal functions, but it has not been reported so far. Efficient repression of E6 has been previously achieved by different methods but, so far, it has not been reported what effect it could have over the expression of mdm2. Different strategies for gene suppressing have been developed over time, amongst which is the use of the relatively new RNA interference. RNAi, known also as post-transcriptional gene silencing, promotes the specific degradation of the target mRNA and such silencing can reach an 85% of effectiveness, therefore, is known as gene knock-down. This kind of efficient and specific effect makes of the RNAi a tempting strategy in searching for the effects that silencing one gene could have over the expression of another associated gene. In the present work the mRNA’s of E6 and E7 (HPV-16) were silenced using RNAi and the results were analyzed by semi-quantitative PCR to determine if the silencing of either oncogene has any effect over the expression pattern of mdm2. It was found that the inhibition of the expression of E6 and E7 has no modifying effect in the expression of mdm2, however, in silencing mdm2 the expression pattern of E6 and E7 is negatively modified whereas the expression of p53 is positively modified.
